For Professor Sherman Klump (Eddie Murphy), being the big man on campus wasn't all it's cracked up to be. He is 400 pounds, has an extremely dysfunctional family, and no love life.

Tired of being the odd-man out, Sherman invents a revolutionary fat gene formula and tests it out on himself. With one swig, the gentle, painfully shy academic is transformed into the lean, overconfident, ladies man, Buddy Love.

With his new looks and alter ego, Sherman has the confidence to pursue the chemistry department's beautiful new professor, Carla Purty (Jada Pinket-Smith). However, Buddy Love faces a large dilemma when he discovers that this long-lasting formula is actually an unstable substance that can wear off at any time.
